Brand names: Everolimus
12.1 Mechanism of Action Everolimus is an inhibitor of mammalian target of rapamycin (mTOR), a serine-threonine kinase, downstream of the PI3K/AKT pathway. The mTOR pathway is dysregulated in several human cancers and in tuberous sclerosis complex (TSC). Everolimus binds to an intracellular protein, FKBP-12, resulting in an inhibitory complex formation with mTOR complex 1 (mTORC1) and thus inhibition of mTOR kinase activity. Everolimus reduced the activity of S6 ribosomal protein kinase (S6K1) and eukaryotic initiation factor 4E-binding protein (4E-BP1), downstream effectors of mTOR, involved in protein synthesis. S6K1 is a substrate of mTORC1 and phosphorylates the activation domain 1 of the estrogen receptor which results in ligand-independent activation of the receptor. In addition, everolimus inhibited the expression of hypoxia-inducible factor (e.g., HIF-1) and reduced the expression of vascular endothelial growth factor (VEGF). Inhibition of mTOR by everolimus has been shown to r

I tried rapamycin after reading [this article](https://www.healthrising.org/blog/2022/07/06/apamycin-resurgence-doctor-chronic-fatigue-syndrome/) on health rising blog, but it made no difference to my symptoms. Many people believe CFS involves neuro-inflammation so I decided to try the closely rela
Hi, I just found an article and the author claims to have been on rapamycin for 4.5 years. This is probably the longest time I have heard of anyone being on rapamycin in an uninterrupted fashio. It is highly informative so I recommend to check it out: [**My Experience With Rapamycin**](https://desmo
I am 7 weeks post liver transplant. My team asked me to join a clinical study which would take me off Tarc and replace it with a medication called Everolimus. This medication is supposedly just as effective as Tarc but less stressful on the kidneys. Does anyone have any experience with this medicati
I graduated in March as an LVN. I started my new and first nursing job about a month ago. I work at a SNF… I love my job so much and enjoy seeing my patients. Got a message from my transplant hospital today informing me that I’ll be covered through June but no guarantees after that. I didn’t even know they were in negotiations! My meds will be available through June but no idea what July will look like. I can’t afford my meds without insurance. I could a
I work at a kinda big pharma plant, and deal with some unpleasant stuff. Our latest batches are mostly bound for the USA, and a minor amount is for other markets. If you happen to use, or have used some of these medications, i'd like to hear if they really help. These aren't all the meds we make, ju
