5 WARNINGS AND PRECAUTIONS Skeletal muscle effects (e
, myopathy and rhabdomyolysis): Risks increase with advanced age (≥ 65), uncontrolled hypothyroidism, renal impairment, and combination use with cyclosporine,or gemfibrozil
The risk of myopathy and/or rhabdomyolysis with statins is increased with concurrent therapy with cyclosporine, erythromycin, fibrates or niacin
There have been rare postmarketing reports of fatal and non-fatal hepatic failure in patients taking statins, including fluvastatin

LOVASTATIN

WARNINGS Myopathy/Rhabdomyolysis Lovastatin, like other inhibitors of HMG-CoA reductase, occasionally causes myopathy manifested as muscle pain, tenderness or weakness with creatine kinase (CK) above ten times the upper limit of normal (ULN)
Myopathy sometimes takes the form of rhabdomyolysis with or without acute renal failure secondary to myoglobinuria, and rare fatalities have occurred
The risk of myopathy is increased by high levels of HMG-CoA reductase inhibitory activity in plasma
The risk of myopathy/rhabdomyolysis is dose related

How can drugs with very short half lives be administered once daily?

Hey P2 here. Going through the dyslipidemia module I noticed that all statins (except fluvastaton IR) are dosed once daily. How can that be adequate for statins with a super short half life like simvastatin (t1/2 2h)? Wouldnt the drug be almost completely out of your system after 10 hours or so? And

Edit!: Correction - I took a 1/3 daily dose of a supplement that contains statins... My title meant to be specifically *prescription* statins... and, the supplement at 0.4% Monacolin K per serving and me only taking 1/3 a serving... that works out to 400mg \* 0.004 / 3 = 0.5mg dose of lovastati
