12.1 Mechanism of Action Crizotinib is an inhibitor of receptor tyrosine kinases including ALK, Hepatocyte Growth Factor Receptor (HGFR, c-Met), ROS1 (c-ros), and Recepteur d'Origine Nantais (RON). Translocations can affect the ALK gene resulting in the expression of oncogenic fusion proteins. The formation of ALK fusion proteins results in activation and dysregulation of the gene's expression and signaling which can contribute to increased cell proliferation and survival in tumors expressing these proteins. Crizotinib demonstrated concentration-dependent inhibition of ALK, ROS1, and c-Met phosphorylation in cell-based assays using tumor cell lines and demonstrated antitumor activity in mice bearing tumor xenografts that expressed echinoderm microtubule-associated protein-like 4 (EML4)- or nucleophosmin (NPM)-ALK fusion proteins or c-Met. In vitro, crizotinib induced apoptosis and inhibited proliferation and ALK-mediated signaling in ALCL-derived cell lines (containing NPM-ALK) at clin

Hello, My husband has ROS1-positive lung cancer with multiple brain metastases. He was treated with Crizotinib, but unfortunately the brain metastases have progressed. We are considering Lorlatinib as the next treatment option, but we have concerns about availability, cost, side effects, and the app
Hi all, I have posted here before a few times about my dad (64M). He was diagnosed with NSCLC adenocarcinoma stage 4 mets to hip on December 2024. He went through 6 rounds of chemo, tumors responded (shrunk a little but still active) and was disease was considered stable. Then he was put on maintena
Hey everyone, I hoped to pose a question. The UK NHS does not use MET amplification as a biomarker for mutation testing. My father, who has Stage 4 non-small cell lung cancer, was put on the conventional treatment regimen, chemo + immunotherapy (carboplatin-pemetrexed-pembrolizumab), because nothi
Hi, I have a rare stage-IV soft tissue sarcoma with mets to the lungs and lymph nodes. Does anyone on here have any experience with crizotinib (brand name in the USA is Xalkori)? I was taking another, similar MET inhibitor (cabozantinib/Cabometyx), but I'm switching due to progression while on cabom
